We currently have a wide selection of libraries for use
with the DupLEX-ATM Yeast
Two-Hybrid system. Our libraries are supplied as 100 µg
of plasmid DNA purified directly from primary bacterial
transformation plates. We do this to save you the time,
effort, and cost of titering and plating and scraping and
purifying DNA from the library yourself. Also, this
procedure does not require another amplification of the
library, so the potential for loss of rare clones is
minimized. In addition to the DNA, we also provide you with
the primary cells in case you do want to amplify the
library. The size range is determined by restriction
analysis of 48 random clones.

Note: First-strand synthesis
of the cDNA was performed using an oligo-d(T)
primer. The libraries marked with an asterisk (*)
have been amplified an additional time, and DNA has
been purified from the re-amplified cells.
To ensure that large (>2.0 kb.)
as well as small inserts are present in our cDNA
libraries, we size fractionate the cDNA into two
pools. The pools are ligated into the vector and
transformed separately. We guarantee the >75% of
the clones in any of the libraries that we
construct contain cDNA inserts. In fact, most of
our libraries have close to 90% of the clones with
inserts.
