pGILDA: pGilda is an expression vector designed for use in the Lex-A-based Yeast Two-Hybrid System. It is primarily used when a particular bait is potentially toxic to yeast and can be used in place f the normal bait expression vector pEG202. The GAL1 promoter allows expression of the LexA-bait fusion protein only when the yeast are grown in galactose medium (with no glucose present). Also, the plasmid contains a CEN (centromere) sequence that maintains the plasmid at only one copy per cell. The LexA-MCS (multiple cloning site)-ADH terminator is exactly the same as that found in pEG202 (see MCS sequence above). The ARS sequence allows for replication of the plasmid in yeast while the HIS3 gene is used for selection in yeast. The AmpR gene is used for selection in E. coli while the colE1 ori allows the plasmid to replicate in E. coli.
